青花菜品種與黑腐病病原交感作用之蛋白質表現

摘  要

  黑腐病(Xanthomoans campestris pv. campestris; Xcc)為十字花科作物重要的病害之一,常影響作物生長和造成產量的損失。本試驗分析市售17個青花菜商業品種對Xcc206抗性檢測,接種兩週後有8個品種對Xcc206表現抗病,罹病率分別為幸運 (3.89%)、Green-Magic(4.25%)、BCS 064(7.00%)、Green Diamond(8.33%)、TI-1120(5.44%)、綠優青花筍(6.67%)、綠帝(11.22%)和盛豐(24.67%),其中慶農50品種對黑腐病原菌的菌株感病最為嚴重(81.11%);至第四週時,僅剩幸運(7.22%)、Green-Magic(12.92%)、Green Diamond(14.58%)和BCS 064(15.00%)表現抗病,大部分品種均對Xcc206呈現感病現象。

  蛋白質體分析研究青花菜抗病品種兩個Green Diamond與幸運及感病品種慶農50與黑腐病菌Xcc206之交互作用蛋白質表現,接種黑腐病原菌Xcc206兩週後之青花菜葉片進行蛋白質二維電泳分析,電泳圖經ImageMasterTM 2D Platinum 5.0軟體分析後,發現有13 個差異表現的蛋白質,挖取這些蛋白質進一步以LC-MS/MS分析和Mascot軟體搜尋,結果spot 77與spot 80為凝集素蛋白質相關之防禦性蛋白質(putative lectin),spot 266與胰蛋白酶抑制劑(trypsin inhibitor propeptide)有關,spot 296與光合電子傳遞和還原調控SAT1折疊機制有關之ROC4(Rotamase cyt 4:peptidyl-prolyl cis-trans isomerase),spot 299為影響光合效率之細胞色素b6/f複合體鐵硫蛋白-2 (cytochrome b6/f complex iron sulfur subunit 2),及一些未知之蛋白質點,並據此推論這些蛋白質與黑腐病原菌具有抗性或感性之關係。

Identifying Interactive Protein of Xanthomonas campestris pv. campestris
 with the Cultivars of Brassica oleracea var. italica

Abstract

   Black rot of broccoli, caused by Xanthomonas campestris pv. campestris(Xcc), is the most important disease of Brassica oleracea var. italica, which causes severe losses in agricultural production around the world.

Of 17 commercial varieties of broccoli screened for their reaction to black rot disease (Xcc206) for two and four weeks, respectively. After inoculaction for 2-weeks, Lucky(3.89%), Green-Magic(4.25%), BCS 064(7.00%), Green Diamond(8.33%), TI-1120(5.44%), Green-Yu broccoli(6.67%), Green-Di (11.22%) and Sheng Feng(24.67%)are highly resistance to the disease; Only Ching-loung 50(81.11%) susceptible of black rot is the most serious. After inoculaction for 4-weeks, Lucky (7.22%), Green-Magic(12.92%), Green Diamond(14.58%) and BCS 064(15.00%) are highly resistance to the disease. All of the most varieties are susceptible.

We have employed proteome to analysis the interaction of Xcc206 with the susceptible and the resistant cultivars of broccoli plants. We have analyzed the proteomic profiling of broccoli plants inoculated with the pathogen Xcc206 for 2 weeks. The extracted proteins from broccoli were analyzed by two dimensional electrophoresis revealing 13 spots showing differential expression.

The 13 spots in gel were excised and analyzed by LC-MS/MS. The proteins were identification analysis with Mascot software. Five of the differentially protein spots were identified as a putative lectin, a trypsin inhibitor propeptide, a Rotamase cyt 4(ROC4) and a cytochrome b6/f complex iron sulfur subunit 2. The putative roles of part these identified proteins in plant susceptibility or resistance relative to black rot disease is discussed.
